Description Introduction: Antibiotics were widely used to cure with infectious diseases, but many people use antibiotics inappropriately, such as not complete and antibiotic use without a prescription, so that the number of resistance increases. To overcome this, another alternative is needed to cure with infectious diseases, namely by combining antibiotics with herbs. This study was conducted with the aim to determine the effect of the combination of sambiloto extract with antibiotic AMX, CHL and COT on S. aureus and E. coli bacteria.Method: The ZOI test was carried out using the AZDAST method with well modification. Observation of ZOI results was carried out by measuring the diameter of the clear zone. ZOI was tested statistically using Kolmogrov-Smirnov, then non-parametric tests using Mann-Whitney were performed. The Minimum Inhibitory Test (MIC) was carried out by microdilution using 96-well plates and macrodilution using a test tube. Microdilution test was observed using a spectrophotometer, while the macrodilution test observed turbidity and the presence of white deposits on the bottom of the test tube. The Minimum Bactericidal Concentration Test (MBC) is a continuation of the MIC test by looking at bacterial growth in agar media. Combined interactions in the MIC and MBC tests are calculated using the Checkerboard assay.Results and Discussion: The combination of sambiloto extract with AMX, CHL, and COT against S. aureus bacteria is additive in the ZOI test. Whereas in the AMX combination of E. coli bacteria, smaller ZOI diameters were obtained, but the statistical test did not show a significant difference. The KBM test cannot be evaluated because it is outside the dose tested.Conclusion: The combination of sambiloto extract with AMX, CHL and COT is additive in the ZOI and MIC tests. However, in these combinations the value of MBC cannot be evaluated because it is outside the test dose.
