Introduction: Malathion has inhibiting effect to the acetylcholinesterase enzyme. It is producing free radicals that can affect the structure and function of respiratory organs. Urena lobata leaves have antioxidant, anti-inflammatory, and anticholinergic effects. This study aims to prove the influence of Urena lobata (DUL) leaves decoction on respiratory frequency and histological features of Danio rerio's gill lamellae, which chronically exposed to malathion.Methods: Adult Danio rerio aged over four months were divided into two control groups and three treatment groups (n=10). Malathion 5 mg/L was given for 40 days in all groups except to the negative control group. DUL was given to the treatment group at dose of 125 mg/L, 250 mg/L, and 500 mg/L. Respiratory rate was analyzed using GOM Player software. Lamellae gills were stained using Hematoxylin-Eosin (HE) and observed under a binocular microscope at 1000x magnification. Data were analyzed using ANOVA test followed by LSD test (p<0,05).Results: Exposure to malathion can increase the frequency of breathing and hyperplasia in the positive control group approximately two times and four times compared to the negative control group (p<0.05). The administration of DUL of 125 mg/L and 250 mg/L inhibits an increase in respiratory frequency of about 20%, while doses of 125 mg/L, 250 mg/L, and 500 mg/L inhibit the gill lamellae hyperplasia of 30%, 50%, and 20%, respectively, compared to the positive controls (p<0.05).Conclusion: DUL dose 250 mg/L is the most effective dose in inhibiting the increase in respiratory frequency and secondary lamellae hyperplasia to gills of Danio rerio.Keywords: Urena lobata, respiratory frequency, gill lamellae, Danio rerio, malathion, chronic
