Introduction: The plants that can be used to treat infections are Imperata cylindrica. Currently, bacteria are resistant to several antibiotics, so it is necessary to act to overcome this resistance with combine medicinal plants with antibiotics. This study aims to determine the interaction of the combination of Imperata with antibiotics against E.coli and S.aureus.Methods: This study uses on in vitro experimental laboratory. Zone of inhibition (ZOI) test was performed using the well-modified AZDAST method. The combination of the minimum inhibition concentration (MIC) was tested by the macrodilution method and evaluated by observing the presence of deposits in the lower part of the tube. The minimum Minimum Bactericidal Concemtration (MBC) is a follow-up of the MIC test performed on a petri dish, and the MIC is removed from each suspension to determine if bacterial growth is present or not.Results: The ZOI combination of cane with amoxicillin (AMX), chloramphenicol (CHL) and cotrimoxazole (CTX) in E.coli significantly increased the diameter of clear zone. The results of the combination with AMX and CTX ZOI in S.aureus were worth the same as their single results, while combinations of CHL with rods significantly reduced the diameter. The results of the fractional inhibitory concentration index (FICI) from MIC values of all antibiotics show additive interactions with both bacteria. The fractional bactericidal concentration index (FBCI) from MBC value indicating synergistic interactions with Imperata and S.aureus and additives to other antibiotics. In E.coli, FBCI was evaluated only with CHL antibiotics with additive values.Conclusion: Imperata increases the effectiveness of the three antibiotics against E.coli bacteria. In S.aureus Imperata only increases AMX and CHL, so it can be concluded that the combination is synergistic. The combination of rods with CHL against E.coli is bactericidal, as is AMX and CTX against S.aureus. In combinations of herbs with S. aureus CHL, it is bacteriostatic.
