Description Introduction: Infectious disease is major problem in Indonesia. The right antibiotic infiltration strategy will reduce incidence rate of infection and resistance. Moreover, resistance can be overcome by increasing antibiotics action by combining it with herbs which have an active antibiotic compound such as Elephantopus scaber Linn. E. scaber is a herb that has already been carried out by research which is found to contain flavonoids, steroids and alkaloid. This research aims to examine the group of polar compound, and examine those compound combinations with amoxcillin and chloramphenicol, measure the inhibition change toward S.aureus and E. coli.Methods : : The Methanol Extract is fractionated with silence phase by using Silica Gel and motion phase by using pure ethylacetate. The interaction between herbs and antibiotics is assessed by Ameri-Ziaei Double Antibiotic Synergism Test (AZDAST) method and the assessment of inhibition is by using Kirby-Bauer method. Phytochemical test is done with formaldehyde spray, FeCL3, and dragendorff of thin layer of chromatography. The result was analyzed by using SPSS with the significant level of p<0,05.Results: Phytochemical test fraction 27 has Alkaloid, Fenol, and Saponin compound while fraction 28-31 has Alkaloid and Fenol compound. Fraction 32 has Alkaloid compound. The combination of antibiotics fraction 27 and 28 has antagonist interaction with chloramphenicol against E. Coli with ZOI values 8,6 +- 8,6 and 9,0 +- 0,0 and significance (p>0,05) while on fraction 30 chloramphenicol against S. Aureus Bacteria has antagonistic interaction with ZOI values 13,6 +- 3,0 and significance (p>0,05). In the combination of amoxicilin antibiotics against S. Aureus and E. Coli obtained type of interaction is not distinguishable.Conclusion: Single herbal F27-32  has no antimicrobial activity. In fraction 27 contains alkaloid, saponins, and phenol, while fraction 28-31 contains alkaloid and phenol.
