Description Introduction: Irrasional use without appropriate indications leads to increased cotrimoxazole resistance. Green tea leaf (C.sinensis) can be used as an alternative treatment of infectious diseases because its contains antibacterial compounds. This study was conducted to define the antimicrobial activity of cotrimoxazole combined with C. sinensis leaf (methanolic extract; decoction) against S. aureus and E. coli.Method: The antibacterial activities (single;combined) were measured through zone of inhibition (ZOI) test with agar well diffusion method. The results of the ZOI combinations were interpreted with the Ameri-Ziaei Double Antibiotic Synergism Test (AZDAST). The data was analysed with Kolmogorov-Smirnov, followed by a non-parametric test Mann-Whitney Test, significant at p<0.05.Results: Combination of Methanolic extract of C. sinensis leaf with Cotrimoxazole (EMCK) and combination of Decoction of C. sinensis leaf with Cotrimoxazole (DCK) were significant increased the diameter of ZOI on S. aureus (p<0.05) I.e 25.67 ± 5.8 mm and 25.67 ± 1.53 mm , respectively, compared to cotrimoxazole low dose (10.33±0.58), methanolic extract of C.sinensis leaf high and low doses (9.67±0.58 mm ; 8.67±0.58 mm) and decoction of C.sinensis leaf all doses (0 mm). Otherwise, both EMCK and DCK combinations was not showed clear zones on E. coli.Conclusion: The combination of EMCK and DCK are synergistic and potentiation, respectively on S. aureus.
