Description Introduction: Physical activity is a planned, structured and repetitive exercise which affects serum leptin levels. During strenuous physical exercise, physical stress can occured leading to various health hazzard. Frequency of physical exercise play a role in determining when physical activity turn into a physical stress which affect serum leptin levels. However, which frequency affects serum leptin levels in unknown. Therefore, this study aims to determine the effect of various frequency of physical stress on serum leptin levels in female rats.Methods: This study was conducted using control group post test only design with 5 groups (n = 6 each). Female rats age 11 weeks old were divided into KNPP (pretreatment group), control (KNP) and various physical stress given daily (KSHN), every two days (KDHN), and twice a week (KSKN). Physical stress were given by forced swimming for 5 minutes without adaptation and were conducted for 4 weeks. At 15 weeks of age, all rats were sacrificed and serum samples were collected. Leptin levels were measured using Rat-ELISA-Leptin Kit and results were analyzed using One Way Anova test followed by post test hoc LSD test and p is set at <0.05.Results: There was no significant difference between pretreatment and negative control suggesting that leptin levels were unchanged in non-exercised groups. KNP (4.41 ± 0.65) was significantly different with KDHN (3.41 ± 0.41, p = 0.003) and KSHN (3.5 ± 0.47, p = 0.007). There is also a significant different between KDHN and KSHN (p = 0.018) suggesting a lower levels of leptin in the serum of excercised animals.Conclusion: Various frequency of physical stress is able to decrease serum leptin levels in female rats subjected to forced swimming
