Introduction: Malathion is a pesticide which has the effect of acetylcholinesterase inhibitors and free radicals which can damage nervous system . Urena lobata water extract has potential as an antioxidant and anticholinergic. This study aims to prove the effect of Urena lobata water extract on brain malondialdehyde (MDA) levels and swimming speed changes in the adult Danio rerio exposed to chronic malathion.Method: Experimental laboratory in vivo with post test only control group design using Danio rerio aged >4 months divided into 2 control groups and 3 treatment groups. Malathion exposure was given for 40 days to all groups except negative control. Urena lobata leaf water extract is given at P1 (125 mg/l), P2 (250 mg/l), and P3 (500 mg/l). MDA levels are observed with MDA Colorimetric Assay Kit Thiobarbituric Acid (TBA) and swimming speed changes were observed with software tracker. Data analysis using one way ANOVA and LSD with a significance level of p <0.05.Result: Urena lobata water extract at doses of 125mg/l, 250mg/l, and 500mg/l inhibited increase of MDA levels by approximately 40%, 70%, and 60% and significantly inhibited decrease of swimming speed changes about 40%, 60%, 20% compared to positive control (p <0.05). The best therapeutic dose of Urena lobata to inhibit the increase in MDA levels and inhibit decrease in swimming speed is 250 mg / l.Conclusion: Urena lobata water extract inhibited increase of brain MDA levels and decrease of swimming speed changes in the Danio rerio exposed to malathion chronically.
