Introduction:Malathion has the effect of acetylcholinesterase inhibitors and free radicals that can affect erythrocyte and Hb level. Urena lobatawater extract has potential as an antioxidant, anti-inflammatory, and membrane stabilisator, but research on the Urena lobata leaf effect to erytrhocyte is still rare. This study aims to prove the effect of Urena lobatawater extract on hemoglobin levels and the percentage of erythrocyteabnormal form in thejuvenile phaseof Danio rerio exposed to chronic malathion.Method:Experimental laboratory in vivo with posttest only control group design using Danio rerio aged 45day divided into 2 control groups and 3 treatment groups (n=5). Malathion exposure was given for 40 days to all groups except negative control. Urena lobata leaf water extract is given at P1 (125 mg/l), P2 (250 mg/l), and P3 (500 mg/l) for 40 days. Hb levels are observed with a comersialHb meter and abnormal erythrocyte images were observed on blood smear with giemsa staining technique then observed in a microscope with 400x magnification. Data analysis using oneway ANOVA and LSD with a significance level of p <0.05.Result: Urena lobata leaf water extract at doses of 125mg/l, 250mg/l, and 500mg/l inhibited Hb levels depressionapproximately 10%, 40%, and20% compared to positive control,while abnormal erythrocytes enhancementinhibited respectivelyabout 40%, 70%, 50% compared to positive control (p <0.05).Chronic malathion exposure decreased Hb level approximately 20% and abnormal erythrocyte percentage 30 fold compared to normal control (p<0,05)Conclusion:Urena lobataleaf water extract inhibited the depressionof Hb level and enhancement oferythrocytesabnormal form in the juvenileDanio rerio exposed to malathionchronically.Keywords:Urena lobata, malathion,erythrocyte, hemoglobin.
