Description Background: Antibiotic resistance in Indonesia continues to increase, so new antibiotics need to be found. Gracilaria verrucosa (Hudson) Papenfus (GV) contains secondary metabolites such as flavonoids, terpenoids, and alkaloids which have antibacterial effects. Solvents in extraction can affect binding of secondary metabolites. However, research on variations in solvents in the extraction process is still limited. Therefore this study aims to look at the effect of variations on the antibacterial activity of GV.Method: GV extraction was carried out by maceration method using 70% ethanol or chloroform variation. The phytochemical test then used reagent solution qualitatively. The antibacterial activity test was assessed through the Inhibition Zone (ZOI) with the well method, the Minimum Inhibitory Concentration (MIC) and Minimum Killer Concentration (KBM) by qualitative macrodilution method in S. aureus and E. coli.Results: The results of phytochemical test of 70% ethanol extract and chloroform found that the active ingredient was terpenoid in 70% ethanol extract. And alkaloids, terpenoids, and flavonoids in chloroform extract. Chloroform extract has significant antibacterial activity in S.aureus bacteria with the largest ZOI value of 11.33 mm and antibacterial activity in E. coli bacteria but not significant. While 70% ethanol extract does not have antibacterial activity on S. aureus and E. coli.Conclusion: : Chloroform solvent Gracilaria verrucosa extract has an antibacterial effect on S.aureus, while 70% ethanol extract does not have an antibacterial effect on S.aureus and E.coli.Keywords: Gracilaria Sp., Antibacterial, S. aureus, E. coli.
