Description Introduction: Diabetes mellitus (DM) is the metabolic disorder with a chronic hyperglycemic condition which can cause microvascular complication. Sambiloto (Andrographis paniculata), Salam (Syzygium polyanthum), Kayu Manis (Cinnamomum verum), and Temulawak (Curcuma xanthorrhiza) is the herbs that have antioxidant dan hypoglycemia effect. Giving a combination of the four herbs can have a stronger effect in the antihyperglycemic mechanism. But, antihyperglycemic mechanism combination Sambiloto, Salam, Kayu Manis, and Temulawak effect on arterial tissue nitric oxide in vivo on hyperglycemia rats model never tested yet.Method: This research was laboratory experimental in vivo control group post test only design. This research used male Wistar rats aged 2-3 months as a animal trial which were divided into 5 groups as samples, positive group, negative group, treatment group 1, treatment group 2 and treatment group 3. Hyperglycemic rats model induce by 10% High Fructose Diet (HFD) and repeatedly low-dose streptozotocin (STZ). Combination extracts of sambiloto, salam, kayu manis and temulawak with a dose of 125 mg / kgBB, 250 mg / kgBB, and 500 mg / kgBB given for 30 days. Nitric oxide tissue was examined using the SKT-2 Total Nitric Oxice Assay Kit. Data analysis used One Way ANOVA (p <0.05).Result: The result showed that the level NO in the KN is signifincantly higher than KP (p<0,05). KP signifincantly different with P2. Giving combination of herbal with a dose of 250 mg/kgBB and 500mg/kgBB able to increase NO levels significantly compared to KP. The higher dose of the herbal combination ther is a tendency to reduce NO levels.Conclusion: Combination of Sambiloto, Salam, Kayu Manis, and Temulawak infusion with a dose of 250 mg/kgBB was able to increase of arterial tissue nitric oxide in male hyperglycemia models of Wistar rats.
