Description Introduction: Antibiotics are used to treat the bacteria infection. However, the irrational use of antibiotics can trigger resistancy. The Herbal combination such as meniran (Phyllanthus niruri) and antibiotics needs to be thoroughly observed because it can be a proper alternative to reduce resistancy. Crude extracts of meniran is known to enhance the effectiveness of antibiotics . However, active compounds that work have not been known yet. This research aimed to assess the interaction of a combination of fractions 24-28 with amoxicillin and chloramphenicol against S. aureus and E. coli.Method: Fraction 24-28 was obtained from methanolic extract process of meniran with12.5 ml of ethyl acetate: 37.5 ml of methanol eluent. Phytochemical tests of the fractions used thin layer chromatography plates sprayed with dragendorff, FeCl3, and formaldehyde reagents. Single fraction inhibition test and combination with antibiotics were conducted by the Kirby-Bauer method and assessed based on the Ameri-Ziaei Double Antibiotic Synergism Test (AZDAST) method.Result: Phytochemical test obtained alkaloids in fractions of 26 and 27, steroids and terpenoids in fractions of 25-28, and phenol compounds in fractions of 24-28. fractions of 24-28 singly had inhibition zone in S. aureus with a diameter ranging from 11-13 mm. However, no inhibitory zones were found in E. coli. The combination of the addition of fractions 24-28 with amoxicillin and chloramphenicol in S. aureus and E. coli are not different significantly.Conclusion: Fraction of 24-28 has antibacterial compounds against S. aureus. The combination of fraction 24-28 with amoxicillin or chloramphenicol, showed the interactionto S. aureus and E. coliisnot distinguishable.Key Words: Amoxicillin, Chloramphenicol, Phyllanthus niruri, L., Phytochemical Tests, Zone Of Inhibition
