Introduction:The incidence of infection and resistance against antibiotics is still a problem in the world, including in Indonesia. Adjuvant therapy is one of the therapeutic choice to increase effectiveness of antibiotic. One of the herbs that is known to have antimicrobial compounds is Elephantopus Scaber, but the group of active compounds that interact with antibiotics is not yet known. This study examined thecombination of fraction semi-polar (F9-F14) Elephantopus Scaberwith amoxicillin or chloramphenicol against S. aureus or E. coli.Method: Methanolic extraction using modified maceration method. Fractionated methanolic extract with silica as stationary phase and mobile phase 45 ml of ethyl acetate and 5 ml of methanol. Phytochemical tests are carried out spray on Thin Layer Chromatography. The inhibitory assessment test by single and combination with the Kirby-Bauer method. Interaction types of Elephantopus scaber combination with amoxicillin or chloramphenicol was assessed based on the Ameri-Ziaei Double Antibiotic Synergism Test (AZDAST) method.Results: The fraction with ratio 90% ethyl acetate eluent and 10% methanol obtained 6 fractions, F9-F14. All of the fractions were contain alkaloids and phenols. Singular test, fractions 9 until 13 had clear zones against S. aureuswith the highest value 8,7±0,6 mm from fraction 9. However, fractions 14 did not have clear zones. While, against E. coli all of fraction did not have a clear zones. In the combination test, fraction 14 decreased the amoxicillin activity againstS. aureus and all of fraction also decreased the chloramphenicol activity against S. aureus.Conclusion: The fraction 9-14 of Elephantopus Scaber contain active compounds of alkaloids and phenols which have antibacterial activity. All of Fraction semi-polar (F9-F14) was antagonistic with chloramphenicol against S. aureus, however amoxicillin activity with fraction 14 also antagonistic against E. coli.Keywords:Elephantopus scaber, amoxicillin, chloramphenicol, adjuvant, combination of herbs and antibiotics
