Description Introduction: Osteoarthritis (OA) is a chronic and degenerative joint disease due to articular cartilagedamage of the synovial joint.Osteoartritis is associated with inflammatory processes and free radical formation. Red ginger(Zingiber officinale var rubrum)and cogon grass(Imperata Cylindrica) had anti-inflammation and antioxidativeproperties. This research aime to study the potential of the combination of red ginger rhizome (Zingiber officinale var rubrum)and cogon grass(Imperata Cylindrica) (RGCG) as an anti OA by measuring the OA rats serum Prostaglandin E2 (PGE2). PGE2 is an inflammatory mediator playing a role in the pain and joints destruction of OAMethods: Male wistar rats aged 4 months divided into 5 groups of each namelynegative control group (NC), positive control group (PC), first treatment with dose 9+40 mg/rat/day (T1), second treatment with dose 18+40 mg/rat/day (T2), and third treatment with dose 36+40 mg/rat/day(T3). Rats were induced with Complete Freund’s Adjuvant (CFA) injected intraarticularry 0.1 ml/rat on right knee joint to make theOA model. TheRGCG combination is given for 10 days. The serum PGE2 level was assessed by PGE2 ELISA Kit. Data analyzed using One Way ANOVA followed by Post Hoc test. The result is significant if p<0.05.Result: The dose of RGCG combination at 18+40 mg/rat/day (2,33 ± 0,20) and36+40 mg/rat/day (2,52 ± 0,33) able to decrease the serum PGE2 level significantly (p<0,05). First treatment with dose 9+40 mg/rat/day (2,55 ± 0,46) also decrease the serum PGE2 level but not significantly (p>0,05).Conclusion:Combination of Red Ginger Rhizome and Cogon Grass dose18+40 mg/rat/day (T2) and36+40mg/rat/day (T3) able to decrease the serumPGE2level of OA rats.Keywords: Osteoarthritis,Red Ginger, Cogon Grass, Prostaglandin E2.
